An anomalous phase transition with a marked rise in specific heat, the isobaric thermal expansion coefficient, and the compressibility coefficient at 62.5 degrees C for an equimolar mixture of 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE) and 1-palmitoyl-sn-glycero-3-phosphocholine (PLPC), in water (34 wt.%) has been shown by differential scanning calorimetry, scanning dilatometry and isothermal compressibility measurements. This transition occurs 15 degrees C above a first-order transition observed in the same system, P-31 and H-2 nuclear magnetic resonance results are consistent with the occurrence of 'defects' in the bilayer in the temperature range between the first and the anomalous phase transitions, It is proposed that conically, PLPC molecules prefer regions with high curvature in the defective bilayer, while DPPE molecules are mostly confined to the flat regions of the bilayers. Seventy-three patients with hematological cancers undergoing allogeneic bone marrow transplantation (BMT) were evaluated for event-free survival (EFS) and toxicity, All received 36 g/m(2) cytosine arabinoside (HDA) and 1200 cGy fractionated total body irradiation (TBI). We assessed the association of EFS and toxicities with the following risk factors: age, gender, diagnosis, initial relapse risk and patient-donor histocompatibility. The EFS probability is 33% at 800 days post-BMT, Twenty-six patients (36%) died of toxicity within 100 days and 14 (19%) have relapsed, EFS was inversely associated with age (P < 0.0001) and initial relapse risk (P = 0.007), The risk of pulmonary (P = 0.023) and hepatic toxicity (P = 0.011) increased with age, Diagnosis other than acute lymphoblastic leukemia (ALL) was a risk factor (P = 0.015) for graft-versushost disease (GVHD); and fewer ALL patients died from toxicity (P = 0.014), The probability of sepsis within 100 days post-BMT correlated (P = 0.007) with initial relapse risk. We conclude: (1) the lower EFS and greater pulmonary and hepatic toxicity associated with increasing age indicate a need for less toxic regimens that maintain high antileukemic efficacy for older patients; (2) the high GVHD and sepsis rates seen in certain categories of patients indicate a need for careful definition of eligibility criteria for this still highly toxic treatment.

Ethanol-inducible cytochrome P450 2E1 (CYP2E1) has been shown to be involved in the metabolism of both ethanol and acetaldehyde, Acetaldehyde, produced from ethanol metabolism, is highly reactive and can form various protein adducts, In this study, we investigated the role of CYP2E1 in the production of a 37-kDa acetaldehyde-protein adduct, Rats were pair-fed an isocaloric control or an alcohol liquid diet with and without cotreatment of YH439, an inhibitor of CYP2E1 gene transcription, for 4 weeks. The soluble proteins from rat livers of each group were separated on SDS-polyacrylamide gels followed by immunoblot analysis using specific antibodies against the 37-kDa protein acetaldehyde adduct, In addition, catalytic activities of the enzymes involved in alcohol and acetaldehyde metabolism were measured and compared with the adduct level, Immunoblot analysis revealed that the 37-kDa adduct, absent in the pair-fed control, was evident in alcohol-fed rats but markedly reduced by YH439 treatment. Immunohistochemical analysis also showed that the 37-kDa adduct is predominantly localized in the pericentral region of the liver where CYP2E1 protein is mainly expressed. This staining disappeared in the pericentral region after YH439 treatment. The levels of alcohol dehydrogenase (ADH) and aldehyde dehydrogenase isozymes were unchanged after YH439 treatment. However, the level of the 37-kDa protein adduct positively correlated with the hepatic content of P4502E1, These data indicate that the 37-kDa adduct could be produced by CYP2E1-mediated ethanol metabolism in addition to the ADH-dependent formation. An account of the resource
Cholesterol 7 alpha-hydroxylase plays a crucial role in cholesterol homeostasis. We investigated the regulation of this enzyme in the hamster, a suitable animal model for studying cholesterol metabolism. DNase I hypersensitivity assay revealed the presence of a hypersensitive region in the proximal promoter. Both negative (bile acids, phorbol esters and insulin) and positive (glucocorticoid hormones) effects were mediated through sequences in the region 318 bp upstream of the ATG codon. All-trans-retinoic acid, cAMP, and LDL did not affect transcriptional activity. These findings show that the hamster cholesterol 7 alpha-hydroxylase gene undergoes a predominant negative regulation, as opposed to the rat CYP7A homologous gene. (C) 1996 Academic Press, Inc.

Mitochondria-derived oxygen-free radical(s) are important mediators of oxidative cellular injury. It is widely hypothesized that excess NO enhances O(2)(center dot-) generated by mitochondria under certain pathological conditions. In the mitochondrial electron transport chain, succinate-cytochrome c reductase (SCR) catalyzes the electron transfer reaction from succinate to cytochrome c. To gain the insights into the molecular mechanism of how NO overproduction may mediate the oxygen-free radical generation by SCR, we employed isolated SCR, cardiac myoblast H9c2, and endothelial cells to study the interaction of NO with SCR in vitro and ex vivo. Under the conditions of enzyme turnover in the presence of NO donor (DEANO), SCR gained pro-oxidant function for generating hydroxyl radical as detected by EPR spin trapping using DEPMPO. The EPR signal associated with DEPMPO/(center dot)OH adduct was nearly completely abolished in the presence of catalase or an iron chelator and partially inhibited by SOD, suggesting the involvement of the iron-H(2)O(2)-dependent Fenton reaction or O(2)(center dot-)-dependent Haber-Weiss mechanism. Direct EPR measurement of SCR at 77 K indicated the formation of a nonheme iron-NO complex, implying that electron leakage to molecular oxygen was enhanced at the FAD cofactor, and that excess NO predisposed SCR to produce (center dot)OH. In H9c2 cells, SCR-dependent oxygen-free radical generation was stimulated by NO released from DEANO or produced by the cells following exposure to hypoxia/reoxygenation. With shear exposure that led to overproduction of NO by the endothelium, SCR-mediated oxygen-free radical production was also detected in cultured vascular endothelial cells. (C) 2011 Elsevier B.V. The effect of 1-palmitoyl lysophosphatidylcholine (PLPC) on the phase behaviour of 1,2-dipalmitoyl phosphatidylethanolamine (DPPE) in excess water (34 wt%) has been examined by differential scanning calorimetry, scanning dilatometry and isothermal compressibility measurements. Mole percentages of PLPC in DPPE between 14 and 62% have been studied over the temperature range 30-75 degrees C. The temperature dependence of orientational ordering at selected sites in H-2-labelled PLPC and 2H(2)O has been determined from measurement of time-averaged chemical shift anisotropies and quadrupole splittings in the P-31- and H-2-NMR spectra. These data have been used to further characterize phase behaviour. At less than equimolar contents of PLPC, when a single phase transition with a reduced transition temperature is observed, spectral and calorimetric data indicate complete miscibility of the two lipid components. An equimolar mixture of PLPC and DPPE shows a sharp first order transition at 47.3 degrees C and a second order transition at 62.5 degrees C. NMR data are consistent with the existence of a defective bilayer at intermediary temperatures. In this range it is proposed that PLPC molecules prefer regions with high curvature in the vicinity of the defects, while DPPE molecules are mostly confined to flatter regions of the bilayer. A possible molecular model is described, At temperatures above 62.5 degrees C, PLPC and DPPE are completely miscible and exist as lamellae. At higher PLPC content (> 50 mol%), thermodynamic and spectral data are indicative of phase separation of the two components over the temperature range examined.

The effect of incorporation of 1-palmitoyl-sn-glycero-3-phosphocholine (PLPC) on the structure of the P-beta' ripple mesophase in aqueous dispersions of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) has been studied by differential scanning calorimetry (DSC) and scanning dilatometry (SD). For samples containing 34 wt. % (H2O)-H-2 and 0-15 wt. % PLPC, a pretransition was observed by DSC. The pretransition disappears at 15 wt. % PLPC. The behavior of thermodynamic functions at the pretransition and main transition gives new insights on the structural changes produced by PLPC on bilayers of DPPC. Endothelial cell polarization and directional migration is required for angiogenesis. Polarization and motility requires not only local cytoskeletal remodeling but also the motion of intracellular organelles such as the nucleus. However, the physiological significance of nuclear positioning in the endothelial cell has remained largely unexplored. Here, we show that siRNA knockdown of nesprin-1, a protein present in the linker of nucleus to cytoskeleton complex, abolished the reorientation of endothelial cells in response to cyclic strain. Confocal imaging revealed that the nuclear height is substantially increased in nesprin-1 depleted cells, similar to myosin inhibited cells. Nesprin-1 depletion increased the number of focal adhesions and substrate traction while decreasing the speed of cell migration; however, there was no detectable change in nonmuscle myosin II activity in nesprin-1 deficient cells. Together, these results are consistent with a model in which the nucleus balances a portion of the actomyosin tension in the cell. In the absence of nesprin-1, actomyosin tension is balanced by the substrate, leading to abnormal adhesion, migration, and cyclic strain-induced reorientation.

The omega-hydroxylation of fatty acids by certain cytochrome P450 enzymes shows a degree of chain-length and regiospecificity which is remarkable in view of the conformational flexibility of these substrates, the strong similarity in properties among homologs, and the lack of polar groups (other than the carboxy terminus) with which to guide and strengthen enzyme-substrate interactions, To investigate the chemical basis for these features of omega-hydroxylation we designed and synthesized a series of lauric acid analogs and evaluated them as substrates and inhibitors of omega-hydroxylation catalyzed by cytochrome P4504A1 and a cytochrome P450 4A1/NADPH-P450 reductase fusion protein, Among n-alkanoic acids, lauric acid was found to have the optimum chain length for the fusion protein, as it does for native cytochrome P450 4A1, With both enzymes, chain shortening caused a precipitous drop in turnover while chain lengthening caused a gradual drop in turnover, The fusion protein omega-hydroxylated methyl laurate and lauryl alcohol about 1/10th as efficiently as lauric acid, but it did not hydroxylate lauramide, 10-Methoxydecanoic acid underwent O-demethylation (via omega-hydroxylation). The branched substrate 11-methyllauric acid was hydroxylated efficiently and selectively at the omega-position, In contrast, the cyclopropyl analog 11,12-methanolauric acid was not detectably hydroxylated, although it induced Type I binding spectrum and inhibited lauric acid omega-hydroxylation by 43% at equimolar concentrations, omega-(Imidazolyl)-decanoic acid induced a Type II heme-binding spectrum and was an especially potent inhibitor of lauric acid hydroxylation, Collectively these data suggest that the active site of cytochrome P450 4A1 has an elongated tubular shape of definite length (ca, 14 Angstrom) with a recognition site for polar groups (including but not limited to carboxyl) at its entrance and the (oxo)heme group at its terminus, (C) 1995 Academic Press, Inc.
